BACKGROUND
Prior studies have shown that the epitope specificity of antibodies influences the clinical presentation of collagen autoimmunity. An epitope is the part of an antigen molecule that an antibody recognizes and binds to. Epitopes are structurally classified as either linear or conformational. Linear epitopes comprise a continuous sequence of amino acids localized in a certain region of the protein sequence, implying that they are close not just in sequence but also in structure. Conformational epitopes, on the other hand, are non-adjacent protein sequence segments that produce a distinctive 3D conformation in a protein's tertiary structure.

Autoantibodies in a patient's serum can react with peptides of a self-protein, the intact self-protein, or both; these autoantibodies may coexist in the serum. Autoantibodies against collagens have been linked to a variety of autoimmune disorders. The discovery of distinct epitope sites on collagens may allow us to determine the role of autoantibodies and different epitope sites in diverse autoimmune disorders. Furthermore, it would pave the way for future diagnostic biomarkers and focused therapy approaches.

THE PROJECT
We are seeking a motivated and enthusiastic master's student to join our research team for an exciting project focused on epitope mapping of type III collagen in autoimmune diseases. Type III collagen is a crucial component of the extracellular matrix and has been shown in various autoimmune conditions, including rheumatoid arthritis, systemic lupus erythematosus, and systemic sclerosis.

In this project, the successful candidate will be involved in the purification of type III collagen autoantibodies from human serum obtained from autoimmune patients. Using biochemical and immunological techniques, including chromatography and immunoassays, the student will isolate antibodies and characterize binding sites within type III collagen that are targeted by autoantibodies in autoimmune diseases.

YOU
We are looking for a highly motivated and detail-oriented Master student with a strong background in biochemistry, immunology, or related disciplines. We are looking for candidates who are passionate about delving into the scientific literature and complementing their laboratory work with a thorough understanding of the theory behind it. Prior experience with protein purification techniques and immunoassays would be advantageous but not required. A good level of English communication skills, both written and verbal, is required, and the ability to work independently as well as part of a team is essential.
