Responsibility and tasks:
  • Design of antibody-based ELISA assays including target identification and design of experiments for generation of monoclonal antibodies targeting the antigen of interest.
  • Purification and characterization of antibodies.
  • Characterization of antibodies including analysis of antibody-antigen interactions and evaluation of specificity and native reactivity by using techniques like ELISA, immunoprecipitation in combination with mass spectrometry, Western blotting and immunohistochemistry.
  • ELISA assay development and method validation including technical, biological and clinical validation. The technical validation included evaluation of parameters like native reactivity, robustness and reproducibility of the method. The biological validation included characterization of the antibodies and antigens in vitro and in situ by using techniques like immunohistochemistry, mammalian cell cultures, Western blotting and SDS-PAGE. Finally, the clinical validation included evaluation of the assays by using samples from clinical cohort studies.
  • - Development of assays using the MSD platform and conversion of existing ELISAs to MSD format.
